背景:
c-Fos is a member of the Fos family of nuclear oncogenes, which also includes FosB, Fos-related antigen 1 (FRA1), and Fos-related antigen 2 (FRA2). As a nuclear phosphoprotein, c-Fos is characterized by its basic leucine zipper (bZIP) domain, which consists of a basic DNA-binding region adjacent to a leucine zipper motif, enabling it to form heterodimers with Jun proteins, thereby constituting the activator protein-1 (AP-1) transcription factor complex. c-Fos contains distinct N-terminal and C-terminal transcriptional activation domains, including motifs such as HOB1 and HOB2, which are essential for its transcriptional activity and oncogenic transformation. Additionally, an extra N-terminal transcriptional activation domain is indispensable for its full transforming potential. c-Fos is classified as an immediate early gene that can be rapidly and transiently induced by a variety of extracellular stimuli, such as growth factors, cytokines, and stress signals. c-Fos regulates the expression of genes related to cell proliferation, differentiation, survival, angiogenesis, and epithelial-mesenchymal transition through AP-1, making it a key regulator in development and tumorigenesis. Phosphorylation by kinases such as Erk enhances c-Fos stability and promotes its nuclear localization, further augmenting its transcriptional activity. c-Fos only forms heterodimers with Jun family proteins (c-Jun, JunB, JunD) and cannot form homodimers, and different Fos/Jun heterodimers exhibit distinct transcriptional activation capabilities. Aberrant regulation of c-Fos expression or phosphorylation leads to tumor transformation and cancer progression.
