背景:
RUNX2 (Runt-related transcription factor 2) is a key transcription factor in skeletal development, primarily known for its role in osteoblast differentiation and bone formation. It acts as a master regulator, guiding mesenchymal stem cells (MSCs) toward the osteoblast lineage by activating key genes that are important components of the bone extracellular matrix, such as type I collagen (Col1), osteocalcin (OCN), and osteopontin (OPN). RUNX2 is essential for both intramembranous ossification (where bone forms directly from mesenchymal tissue) and endochondral ossification (which involves the replacement of cartilage by bone). During endochondral ossification, RUNX2 regulates the maturation of hypertrophic chondrocytes and promotes the expression of type X collagen (Col10a), facilitating the conversion of cartilage to bone. Furthermore, RUNX2 also influences the production and mineralization of bone matrix during osteoblast maturation. It plays a significant role in regulating osteoclastogenesis by modulating genes such as osteoprotegerin (OPG) and RANKL, maintaining the balance between bone formation and bone resorption, thereby preserving bone homeostasis. It is also involved in angiogenesis by regulating vascular endothelial growth factor (VEGF), promoting blood vessel formation in bone tissue. Dysregulation or mutations of RUNX2 can lead to various skeletal disorders, including cleidocranial dysplasia (CCD), osteoporosis, and osteosarcoma.
