背景：
Platelet/endothelial cell adhesion molecule-1 (PECAM-1), also known as CD31, is an important cell adhesion and signaling molecule primarily present in endothelial cells, circulating platelets, monocytes, neutrophils, and some T cells. PECAM-1 is enriched at endothelial junctions, where it plays a crucial role in maintaining vascular integrity by promoting homophilic interactions that support vascular permeability and leukocyte migration. The cytoplasmic domain of CD31 contains immunoreceptor tyrosine-based inhibitory motifs (ITIMs), which, upon phosphorylation by Src family kinases (particularly at Tyr686), recruit various signaling adaptor molecules such as SHP-2, PI3 kinase, and PLC gamma. This phosphorylation regulates signaling pathways that enhance endothelial cell motility and survival. It also positions CD31 as a central scaffold integrating multiple signaling pathways involved in cell adhesion, migration, and angiogenesis.
