背景：
COL1A1 is the gene encoding the pro-alpha 1 chain, which is part of the triple-helical structure of type I collagen. This collagen is composed of two alpha 1 chains and one alpha 2 chain. COL1A1 contains three conserved domains - the von Willebrand factor type C (vWFC) domain, the collagen triple helix repeat, and the fibrillar collagen C-terminal domain (COLF). The expression level of COL1A1 is tightly regulated through multiple mechanisms, such as mRNA stability, translation, and post-translational modifications. Overexpression of COL1A1 can lead to fibrotic diseases in various tissues, such as systemic sclerosis, while loss-of-function mutations in the COL1A1 gene can cause osteogenesis imperfecta, commonly known as brittle bone disease. It also plays a role in regulating cell expansion and the cell cycle in bovine cumulus cells. COL1A1 can also affect apoptosis by regulating the caspase-3 and BCL-2/BAX ratio. It is also associated with osteoporosis and chronic liver injury. Downregulation of COL1A1 can inhibit the proliferation, invasion, and mitosis of human oral squamous cell carcinoma. Activation of COL1A1 can also inhibit apoptosis in cervical cancer cells.
