背景：
Ki-67 is a large nuclear and nucleolar protein encoded by the MKI67 gene, widely used as a marker of cell proliferation. Its structure includes an N-terminal forkhead-associated (FHA) domain, a protein phosphatase 1 (PP1) binding region, central tandem repeats, and a C-terminal leucine/arginine-rich chromatin-binding domain, and it produces multiple isoforms through alternative splicing. Ki-67 is expressed in all active phases of the cell cycle (G1, S, G2, and M phases) but is absent in quiescent cells (G0 phase), making it a reliable proliferation indicator in cancer diagnosis. Functionally, it plays key roles in organizing interphase heterochromatin, assembling the perichromosomal layer (PCL) during mitosis to prevent chromosome aggregation, and regulating cell cycle progression through interactions with PP1 and other chromatin-associated proteins.
