背景：
LAMP-1 (lysosomal-associated membrane protein 1) is an indispensable glycoprotein in the lysosomal membrane of eukaryotic cells, playing a crucial role in maintaining lysosomal integrity and function. Structurally, it is a type I transmembrane protein characterized by a large luminal domain with extensive glycosylation, forming a protective glycocalyx that shields the lysosomal membrane from degradation by acidic hydrolases. This glycosylation protects the lysosome and facilitates interactions with other proteins involved in lysosomal trafficking and autophagy. LAMP-1 participates in chaperone-mediated autophagy (CMA), which aids in the selective degradation of cytosolic proteins, and plays a key role in sorting lysosomal enzymes to appropriate destinations. Additionally, LAMP-1 is expressed on the surface of various immune cells, contributing to the regulation of immune responses and preventing self-damage during cytotoxic activities. Dysregulation or deficiency of LAMP-1 can lead to lysosomal storage disorders and may contribute to cancer progression and immune evasion.
